Design Notes

An important insight into the design of the construct came from the expertise of Prof. Dr. Mario Moerl, who emphasized the importance of maintaining the natural environment of the riboswitch (BBa_K4654011). This is crucial because it influences the folding and functionality of the riboswitch. We retained the spacer sequence containing the ribosome binding site (RBS) and the 5' region of the nhaA gene between the riboswitch and the reporter sequence. For this, we used the same sequence configuration that White et al. (2022)1 used in their experiments.

The sequence we are using was created by White et. al. by creating a consenus sequence from different sequences of the riboswtich family1.
